I have updated some of the wikis:
http://qt-project.org/wiki/Get_The_Source
and
http://qt-project.org/wiki/Building-Qt-5-from-Git
The problem in the first wiki was that it pointed to the 4.8 docs.
the README file you mentioned is also updated and will soon (tm) land.
This duplication of information is not ideal (in addition we have the Qt docs) but I'm not sure of how to fix it properly.

Hii,
there isn't any information about this in qt-everywhere-opensource-src-5.0.0-rc1.zip and there is no requirements-win.html file in the source package.
The README in the top directory mentions the system requirements perl, python and c++ compiler for C++98. It also contains a link to http://qt-project.org/wiki/Get_The_Source.
I did not find anything about ruby there. A web search for ruby site:q-project.org/doc/qt-5.0 also did not find anything useful.

According to the docs in the binary package, Ruby should be documented:
C:\qt\Qt5.0.0-rc1-368\5.0.0-rc1\msvc2010\doc\qtdoc[x86]> grep ruby *
requirements-win.html:<li>Install <a href="http://www.ruby-lang.org">Ruby</a> from <a href="http://rubyinstaller.org/">here</a> and add the
installation location to your PATH in order to be able to build QtWebKit.</li>
Can you double-check this?

There are still problems with compiling qt-everywhere-opensource-src-5.0.0-rc1.zip with VS2010:
- ruby is required (undocumented)
- "configure" with "nmake" fails when building module-qtwebkit-examples-and-demos with:

- "configure -nomake examples" tries to build module-qtwebkit-examples-and-demos
- Documentation is not built (with the exception of assistant.qch)
